COMBINED EFFECTS OF rHu GM-CSF AND rHu-IFNalfa-2b ON INTERFERON - SENSITIVE OR RESISTANT LYMPHOID CELLS

Background: The treatment of cancer with cytostatic drugs is one of the three major ways to confront this unusual proliferation of cells. Before any use of new drugs or combined use of well known drugs in patients, some experiments take place in cancer cell lines. From this point of view four lymphoid cell lines, established from BurkittOs lymphoma, have been treated with IFNalfa-2b and GM-CSF, two biological modifiers which are in use as antiproliferative drugs. The synergistic effects of these two cytokines have been studied by measurement the induction of 2'5' oligo (A) synthetase. Methods: Four lymphoid cell lines have been used in our experiments. They were grown as stationary suspension and different concentrations of IFNalfa-2b and GM-CSF were added to examine the synergistic effect of these two cytokines. The activity of 2'5' oligo (A) synthetase was estimated by following the conversion of 3H-labeled ATP to adenosine oligomers as previously described by the authors. The activity of the enzyme is expressed as the percentage of the substrate which is converted to a mixture of oligonucleotides. Results: It is obvious that the GM-CSF has no any effect on the proliferation of the four lymphoid cell lines after three days incubation. However there is an obvious synergistic effect of the two cytokines on the proliferation of the cells and on the induction of the 2'5' oligo (A) synthetase. Conclusion: A possible mechanism of interaction between IFNalfa-2b and GM-CSF could explain the synergistic effect on the proliferation of the four cell lines. The results obtained and mainly the increase of the sensitivity to IFNalfa-2b, suggest the antitumor effect of Ganulocyte-Macrophage Colony Stimulating Factor, an observation which seems to be more true now than some years before. |
