  OXIDATIVE STRESS AND GLUTATHIONE - ASSOCIATED ANTIOXIDATIVE SYSTEMS IN MALIGNANT CELLS

  Abstract:
  Free radical-mediated damage appears to be of primary significance in aging, cancer and many other pathological conditions. Reactive oxygen species are continually produced during the biochemical aerobic reactions, which occur as a part of a normal cell metabolism, or during the biotransformation of xenobiotics, including carcinogens as well as antitumor drugs. Numerous forms of cellular damage are due to oxidative stress induced by free radicals. Glutathione (GSH) is universal protectant against electrophilic challenge and it is essential in the detoxification of redox active drugs. Glutathione is also involved in biochemical mechanisms that can cause tumors to become resistant to anticancer drugs. The importance of glutathione and associated enzymes in antioxidative defense of malignant cells lies in the provision of an enhanced rate and extent of detoxification, helping to maintain the amount of damage below that which may trigger apoptosis.
